If I create an ISO image of my existing Windows 10 WinToUSB disk can I boot to this from a multiboot setup such as YUMI?

I am very sorry, we are not familiar with YUMI. Do you mean you want to start Windows 10 from an ISO? This seems impossible, because Windows cannot be booted from a read-only device.
